Hewlett-Packard (Agilent) 1100 LC-MSD

Our high performance liquid chromatography system is equipped with an in-line solvent degasser, two-channel gradient pump, 96 sample auto-injector, 2 column oven, UV-Vis DAD, and quadrupole mass spectrometer. We have both the Electrospray Ionization (ESI) and the Atmospheric Pressure Chemical Ionization (APCI) interfaces.

Instrument Reservations

You must note times you are using on the web calendar. This is so that users from across campus won't have to walk all the way to the lab to find out it is in use.

Loop Injection of Samples

If you do not need to chromatographically resolve the components of your sample, or you have a nearly pure sample you can bypass the column to get an ESI-MS or APCI-MS spectrum of your sample directly.

Reverse Phase Chromatography

We have various LC columns of various lengths, diameters, particle size, and phase available for public use. Most of these columns can handle solvent conditions from 20% to 100% organic. You should consult the manufacturers' websites or the published chemical literature for assistance in determining what column phase will effect the separation you desire.

When selecting buffers for use with ESI-MS or APCI-MS be sure to choose volatile buffers. Non-volatile buffers like sodium phosphate will deposit salt inside the ion source, reducing the efficiency of ion production.

The EAC provides basic solvents including methanol, acetonitrile, and ultra-pure water. You must provide other solvents or buffers.

Diode-Array Detection UV-VIS Spectroscopy

The UV-VIS detector is equipped with a high-pressure, high-sensitivity cell from the manufacturer. Because most users are not actually using the the UV-VIS data, the detector is normally kept disconnected from the path of solvent flow. If you need the UV-VIS data, contact EAC staff for training in connecting and disconnecting the UV-VIS.

Mass Spectrometer

The mass spectrometer is a quadrupole design with a mass range to 2,000 Daltons. No MS-MS is possible on this instrument, however the ion kinetic energy can be varied through the interface region allowing for greater or lesser degrees of fragmentation.
